GRAND ROUNDS PRESENTATION

"What cell-specific transcriptomes tell us about prostate cell lineages
and how prostate cancer progresses"
 

On Friday October 29, 2004, Lawrence True, MD, Professor of Pathology, Department of Pathology, University of Washington Medical Center, Seattle, WA, presented a continuing medical education lecture on prostate cancer research. Dr. True summarized his research as follows:

We have taken a cell type-specific approach to investigate prostate biology. Based upon CD phenotyping the prostate ( Am J Pathology 160:37-43, 2002), we have sorted prostate parenchymal cells to determine their transcriptomes by expression microarrays. We complemented this approach with laser microdissection and separation of the “side population” of putative precursor/stem cells. Application of these approaches and of normal cell transcriptomes to prostate cancer have enabled us to characterize the transcriptomes of the common Gleason patterns of tumor. Immunohistochemical verification of expression of genes differentially expressed by prostate cancers of different grade has revealed patterns of CD expression by tumor that correlate with tumor aggressiveness, i.e. overexpression of CD10/CALLA. Extending these findings to our Rapid Autopsy metastatic samples has revealed the magnitude of one future challenge, namely, dealing with the degree of heterogeneity of expression of such important tumor phenotypes as the neuroendocrine cell population in the different metastases in a given patient.
